What is the dosage of Pemigatinib?

Pemigatinib is available as an oral tablet in the following doses:

• 4.5mg
• 9 mg
• 13.5 mg

How should Pemigatinib be taken?

The recommended dosage is 13.5 mg orally once daily for 14 consecutive days followed by 7 days off therapy in 21-day cycles.
Treatment should be given until disease progression or unacceptable toxicity occurs. It should be taken orally with or without food.

Missed a Dose?

If a planned dose of Pemigatinib is missed by 4 or more hours or if vomiting occurs, resume dosing with the next scheduled dose.

What are the warnings and precautions for Pemigatinib?

• Pregnant and lactating women should not use this drug

• Safety of usage was not known yet in children

• No dose adjustment is recommended for patients with mild or moderate kidney or liver problems

What are the side effects of Pemigatinib?

•General: Hyperphosphatemia- excess levels of phosphate, tiredness, backpain
•Digestive: Nausea, vomiting, diarrhea, abdominal pain, decreased appetite
•Skin: Nail toxicity, dry skin
•Eye: Dry eyes

What are the other precautions for Pemigatinib?

•Perform a detailed eye assessment before starting Pemigatinib and every 2 months for the first 6 months and every 3 months thereafter during treatment.
•In case of vision problems like blurred vision - refer patients for ophthalmologic evaluation urgently, with follow-up every 3 weeks until resolution of symptom
•The drug may cause dry eyes.
•The drug may increase the levels of phosphate. So phosphate levels to be monitored regularly.
•Female patients with reproductive potential should use effective contraception during treatment and for 1 week after the final dose.
•Males with female partners of reproductive potential should use effective contraception during treatment with PEMAZYRE and for 1 week after the final dose.

What are the Drug Interactions of Pemigatinib?

Pemigatinib should not be given along with inhibitors of CYP3A4.
Reduce dose from 13.5 mg to 9 mg and reduce dose from 9 mg to 4.5 mg in case of concomitant administration with inhibitors of CYP3A4.
