Pexidartinib is an anticancer drug that is used for the treatment of adults with tumors of joint called tenosynovial giant cell tumor who don’t respond to surgery.

Why is Pexidartinib Prescribed? (Indications)

Pexidartinib is an anticancer drug indicated for the treatment of adult patients with tumors of joint called tenosynovial giant cell tumor who don’t respond to surgery.
Symptomatic tenosynovial giant cell tumor is a rare group of cancer tumors that occur in the joints. These types of tumors are typically not cancerous but they can grow and damage the surrounding structures.
Symptoms include pain, swelling, and limitation of movement of the joint. They can also cause significant disability.

What is the dosage of Pexidartinib?

The dose recommended is 400 mg orally, twice daily. The dose should be reduced in kidney disease. They are available as 200mg Capsules

How should Pexidartinib be taken?

Take Pexidartinib on an empty stomach or at least 1 hour before or 2 hours after a meal or snack.
Do not break, open, or chew Pexidartinib capsules.
Swallow the capsules whole.

Missed a Dose?

In case you vomit or miss a dose of Pexidartinib then they should take the next dose at its scheduled time.

What are the side effects of Pexidartinib?

• Before taking Pexidartinib, inform your healthcare provider if you are suffering from liver diseases.
• Pexidartinib can cause serious liver injury. Monitor liver function test before starting Pexidartinib and during the treatment at specified intervals. In case of severe liver toxicity, stop or reduce dose of Pexidartinib.
• During treatment with Pexidartinib, inform your healthcare provider immediately if you develop any symptoms of liver disease like- loss of appetite, vomiting, abdominal pain, fever, rash, itching, nausea, tenderness, fatigue.
• Stop taking Pexidartinib and inform your healthcare provider if you develop symptoms like yellowing of skin, eyes and dark urine.
• Pexidartinib is available through a restricted program because of its risk of hepatotoxicity.
• Embryo-fetal toxicity- Pexidartinib can cause fetal harm when administered to pregnant women.
• Females should use effective contraception during treatment and 1 month after the final dose.
• Advise males with female partners of reproductive potential to use effective contraception during treatment and 1 week after the final dose.
• Do not take Proton pump inhibitors (Pantoprazole, omeprazole, rabeprazole, lansoprazole), St John Worts and grapefruit juice along with Pexidartinib.
• Inform your healthcare provider about all the prescription medicines, vitamins, OTC products and herbal medicines.

What are the Drug Interactions of Pexidartinib?

• Do not use Pexidartinib with other hepatotoxic drugs like allopurinol, amoxicillin, amiodarone, isoniazid, phenytoin, sulfasalazine, etc.
• Do not use Pexidartinib with Phenobarbital, phenytoin, rifampin, glucocorticoid, carbamazepine, etc.
• Do not use or reduce dose of Pexidartinib with amiodarone, cimetidine, erythromycin, sulfonamides, ketoconazole, grape fruit, etc.
• Do not use or reduce dose of Pexidartinib with silybin, atazanavir, quinidine, diclofenac.
• Do not use with proton pump inhibitors- (Pantoprazole, omeprazole, rabeprazole, lansoprazole
